双链DNA结合与打开动力学研究方面取得新进展

传统的反应动力学以两态模型为基础解释寡聚核酸杂交与解链的实验现象。但是,其详细过程显然应该以一个一个碱基的方式进行。化学生物学研究部赵新生教授课题组巧妙地将荧光共振能量传递(FRET)的效率随距离的变化用碱基解开的个数表达,应用单分子水平的荧光相关光谱(FCS)详细探测了这一过程,得到一个一个碱基结合和打开的热力学和动力学数据,描绘出双链DNA解链开始时沿反应坐标的势能变化。这一成果发表在J. Am. Chem. Soc. 2008, 130, 16947一文中。


Traditionally, the kinetics of hybridization/dehybridization of oligonucleotides were treated by two-state models. Clearly, however, the true process should goes in a base-by-base manner. Prof. Xin Sheng Zhao and his coworkers investigated the details of the process by fluorescence correlation spectroscopy (FCS). They labeled a double stranded DNA with dyes capable of fluorescence resonance energy transfer (FRET) and related the number of dissociated bases to the FRET efficiency. Thermodynamic and dynamic information on single base pair reaction was extracted. The base-by-base picture was depicted along the reaction coordinate. The results were published on J. Am. Chem. Soc. 2008, 130, 16947.
